UHM splicing factors functions and perturbations by small molecules
Gene expression requires many layers of regulation. Among which splicing is an essential step consisting in removal of non-coding sequences from pre-mRNAs. Dynamic mediation of protein-protein and protein-RNA interaction are accomplished by ribonucleoproteins in a stepwise manner. One of the splicing factors, U2 small nuclear ribonucleoprotein (snRNP) auxiliary factor (U2AF65) is involved in early spliceosome assembly. The U2AF homology motif (UHM) domain is dedicated for protein-protein interaction to recognize U2AF ligand motifs (ULM) in splicing factors, including SF3b155. We are trying to understand the function of UHM-ULM interaction, and discover small molecules as interactors of UHM domains for further therapeutic purposes.
