Exploring the common and specific functions of UHM splicing factors (UHMSFs)
Pre-mRNA splicing is an essential step in gene expression in higher eukaryotes. Actually most genes are initially transcribed as pre-mRNAs that contain intervening sequences (introns), as well as expressed sequences (exons). For proper expression of proteins, introns that interrupt the coding sequences have to be removed from pre-mRNAs and exons to be joined together during the process of pre-mRNA splicing. Although the splicing reaction is chemically simple, splicing of pre-mRNA requires a diversity of proteins and small RNAs to ensure the “single nucleotide precision” of the reaction so that only functional mRNAs are produced. Splicing defects are known to be a common cause of genetic diseases and recurrent mutations of splicing factors involved in 3’ splice site recognition have been unraveled in a diversity of myelodysplastic syndromes and tumors. In this context, we are taking part in the characterization of interactions that occur during spliceosome assembly using functional and structural approaches. Our studies mainly focus on the mechanism of recognition of the 3’ splice site.
